[Using Sakai] How to create a URL reference to an assignment in a course?

Yep

/direct/assignment/site/siteid.format gives you a list of assignments for the site. That's for trunk and was part of project Keitai. Search jira and there is a commit or two you can pull back to other versions. The URL API is documented in that jira too.
